Using TTP Translational Technology®, TTP435 was identified as a potent and selective inhibitor of AgRP.  TTP435 has been assessed in a series of in vitro and in vivo studies as proof of concept to demonstrate the value of inhibiting AgRP as a safe and effective treatment of obesity.  TTP435 has no effect on MC4R alone or in the presence of αMSH. 

In vivo, TTP435 is orally bioavailable with high brain penetration.  In multiple animal models of obesity ranging in length from overnight administration to 4-week treatment, TTP435 was shown to reduce food intake and body weight gain, reduce fat composition, and reduce insulin levels in a dose dependent fashion.

TTP435 is presently being assessed in obese subjects in phase II clinical trials.
